Summary

Growth hormone and its synthetic analogs have a great influence on plant growth, but different concentrations produce different physiological effects. The effects of growth hormone analogs, one Cai acetic acid (NAA), on root and shoot growth were observed by bioassay, thus providing a theoretical basis for the use of Kiss acetic acid for seed dipping or seed dressing before sowing in the field.

Principle

Solution:
Weighed 10 mg of turbulent acetic acid, first dissolved in a small amount of ethanol, and then reconcentrated to 100 mL with evaporated water to form 100 mg - L

Kiss acetic acid solution, store this solution in the refrigerator and dilute 10 times when used.
Equipment.
Constant temperature incubator, Petri dish, 1 mL, 2 mL, 10 mL pipettes, round filter paper (diameter is the same as the inner diameter of the bottom of the Petri dish), pointed inlays, marker pen.

The basic procedure for the effect of growth hormone analogs on root and shoot growth can be divided into the following steps:

1. 7 Petri dishes were washed and dried and numbered, 10 mL of 10 mg-L-1 NAA solution was added to Petri dish No. 1, 9 mL of distilled water was added to each Petri dish No. 2-6, and then 1 mL of 10 mg-L-1 NAA solution was pipetted from Petri dish No. 1 and injected into Petri dish No. 2, and then 1 mg-L-1 NAA solution was mixed well. Then 1 mL from No.2 dish was sucked into No.3 dish and mixed well to form 0.1 mg - L-1 solution, and then continued to be diluted to No.6 dish, and the result was that the concentration of NAA from No.1 dish to No.6 dish was 10 mg - L-1, 1.0 mg - L-1, 0.1 mg - L-1, 0.01 mg - L-1, 0.001 mg - L-1, 0.000 1 mg - L-1, and finally, 1 mL of NAA solution was sucked from No.6 dish and discarded. Each dish was, 9 mL of solution. dish No. 7 with 9 mL of distilled water was used as control.

2. About 200 wheat seeds were selected, surface sterilized with saturated bleach supernatant for 20 min, removed and rinsed with tap water, and then rinsed three times with evaporated water, and the water on the seed surface was sucked out with filter paper.

Put a piece of filter paper in each Petri dish from 1 to 7, place 20 seeds neatly along the periphery of the Petri dish so that the embryo faces the center of the Petri dish, cover it and place it in the incubator at 20~25 °C for 24~36 h. Observe the germination of the seeds, and leave 10 seeds that have germinated neatly . 3 days later, determine the number of roots, the length of root, and the length of shoots of the seeds of the various treatments, and calculate the average value of them, and record them in Table 21-1 to determine the effect of NAA on the growth of roots and shoots, or the effect of NAA on the growth of roots and shoots. The concentration of NAA that promotes or inhibits the growth of roots and shoots was determined in Table 21-1.

Caveat

1. Wheat seeds should be selected as mature, full and uniform in size.

2. Seeds of uniform growth should be selected for measurement.
