Experimental effects of drugs on blood pressure and heart rhythm in rabbits

Summary

Epinephrine alone has a pressor effect, while alpha-blockers can reverse its pressor effect. Barium chloride can promote the inward flow of sodium ions in the Purkinje fibers of the heart, inhibit the outward flow of potassium ions, promote the 4-phase automatic depolarization, and enhance the autoregulation, so it is commonly used in the manufacture of various types of cardiac arrhythmia models. Lidocaine belongs to the IB class of anti-tachyarrhythmic drugs, which can selectively act on Purkinje fibers, inhibit sodium ion in-flow, promote potassium ion out-flow, reduce autoregulation and eliminate refractory excitation. Clinically, it is often used as the first choice for prevention and treatment of acute myocardial infarction and ventricular arrhythmia. Ventricular arrhythmia of choice.

Move

1. Anesthesia and fixation: One rabbit was taken, weighed, anesthetized with sodium pentobarbital 30 mg/kg intravenously at the ear margin, and fixed in the supine position on the rabbit platform.

2. Surgery: longitudinal incision of the skin in the middle of the neck, blunt separation of the subcutaneous tissue, exposure and separation of one side of the common carotid artery about 2 cm, from the common carotid artery under the two threads, the edge of the ear intravenous injection of heparin 1 ml/kg, ligature distal end, the proximal end of the proximal end of the arterial clip, with ophthalmic scissors towards the proximal end of the direction of the cut small mouth, and then intubation connected to the pressure transducer, ligation and fixation, release the arterial clamp, record the rabbit's normal blood pressure

3. Electrocardiogram: According to the requirements of the instrument, insert the needle electrodes into the subcutaneous part of the limbs of the corresponding leads. Left lower limb, right lower limb ground), record the normal ECG II lead graph.

4. Drug administration: 0.001% Adr 0.1 ml/Kg was administered intravenously from the ear margin to observe the changes in the blood pressure of the rabbits; after the blood pressure returned to normal, 0.001% isoproterenol 0.1 ml/Kg was administered to the rabbits to observe the changes in the blood pressure and to compare the effects of the two drugs on the blood pressure.

After the blood pressure returned to normal, 0.2% barium chloride 2 ml/Kg was given to the rabbits to observe the changes in the heart rhythm, and after the arrhythmia appeared, 0.5% lidocaine 1 ml/kg was given intravenously immediately, and the injection was completed in 1~2 minutes, and the changes in the heart rhythm were observed, and the observation was continued for 5 minutes.

Edit the experimental records, print and analyze the results.

Caveat

1. Administer sodium pentobarbital. Barium chloride should be pushed slowly.

2. arterial cannulae should be securely ligated.

3. Lidocaine should be given immediately after the rabbit develops arrhythmia.

4. electrodes should not be inserted into muscles to avoid myoelectric interference.
