Summary

The TPS is an open gas circuit system for the simultaneous determination of photosynthetic and respiration rates and transpiration rates in plant leaves. When measuring, the leaf is placed in a closed leaf chamber with illumination (if the measurement of respiration does not require illumination), then the instrument automatically detects the gas flow through the leaf chamber, the concentration of CO2 and H2O in the gas entering the chamber, and the concentration of CO2 and H2O in the gas exiting the chamber, and then it automatically calculates and gives the photosynthetic and respiration rates and transpiration rate of the plant leaf.

Principle

The TPS is an open gas circuit system that can simultaneously measure the photosynthetic rate and respiration rate of plant leaves as well as the transpiration rate. When measuring, the leaf is placed in a closed leaf chamber with illumination (no illumination is needed for respiration), then the instrument automatically detects the gas flow through the leaf chamber, the CO2 and H2O concentration in the gas entering the chamber, and the CO2 and H2O concentration in the gas flowing out of the chamber, and then it automatically calculates and gives the photosynthesis rate, respiration rate, and transpiration rate of the plant leaf.

1. Instrument installation

Connect the leaf chamber to the main unit (TPS) and the gas line (R, A) to the interface (R, A) on the main unit correspondingly.

2. Measurement

2.1 Instrument start-up

Toggle the OFF/ON switch to ON. The menu shown on the display is the main menu:

2.2 Select the part of the middle section of the plant leaf where the margins on both sides of the leaf are relatively straight and clip it into the leaf chamber, and measure the width and length of the leaf clipped into the leaf chamber to find out the area of the leaf being measured ( cm2 ).

2.3 Press the number key 1 to enter the measurement mode:

2.4 Press numeric key 2 to select universal leaf chamber and then enter the state of selecting recording type:

2.5 Press numeric key 1 to select M (manual recording), then press letter key Y to confirm and enter the state of selecting light source:

2.6 Determination of photosynthetic rate: press numeric key 1 to select SUN (natural light), if the determination of respiration rate, the light-transmitting surface of the leaf chamber will be covered with black cloth or red paper, and then press numeric key 2 to input the leaf area of the leaf to be measured, and then press the letter key Y to enter into the state of setting the record number:

2.7 Students can not change the record number in the experiment, directly press the letter key Y to enter the measurement state, and wait for the equilibrium for 2min, then start to record the measurement data, and record the readings in Table 1:

2.8 Press the conversion key X that shows the calculation results, every 2min read a number of times, record three readings and seek the average value, the results of the experiment will be recorded in Table 2:

In the measurement state press the number keys 1/2/3 to return to the setup menu; 1 for changing the recording mode; 2 for changing the light source or leaf area; 3 for changing the plot number. After modification, press the Y key to return to the measurement state.

Note: After measurement, you must press N to return to the main menu to turn off the power. Toggle the OFF/ON switch to OFF.
