Product Description

One unit of Vaccinia DNA Topoisomerase I catalyzes the conversion of 0.5µg of supercoiled pUC18 DNA to a relaxed state in 30 min at 37°C.


Application

Vaccinia DNA Topoisomerase I can be used to relax the DNA supercoils for subsequent molecular biological reactions such as enzyme digestion, ligation between DNA vectors and PCR amplified fragments, nick repair and adapter ligation.Definition of activity: One unit of Vaccinia DNA Topoisomerase I catalyzes the conversion of 0.5µg of supercoiled pUC18 DNA to a relaxed state in 30 min at 37°C.10X Reaction Buffer: 500mM Tris-acetate, 1M NaCl, 25mM MgCl2, 1mM EDTA, pH 7.5 at 25°C.Enzyme storage buffer: 50mM Tris-HCl (pH 7.5), 1mM DTT, 0.1mM EDTA, 0.1M NaCl, 0.1% Triton X-100 and 50% (v/v) glycerolInactivation or inhibition: Vaccinia DNA Topoisomerase I can be inactivated by incubation at 80℃ for 20 minutes.

Inactivation or inhibition

Vaccinia DNA Topoisomerase I can be inactivated by incubation at 80℃ for 20 minutes.


Precautions

When supercoiled plasmids are treated with Vaccinia DNA Topoisomerase I, they should be separated on an agarose gel without ethidium bromide (EB). DNA staining with EB or other dyes must be performed after electrophoresis, as EB binds between the bases of DNA and catalyzes the formation of supercoils. After the electrophoresis is completed, the electrophoresis position of DNA fragments will not be changed even if the supercoil is formed again.This product is for R&D only. Not for drug, household, or other uses.For your safety and health, please wear a lab coat and disposable gloves during the operation.


Instructions for Use

1. Thaw all reagents required for the reaction. Mix well and place Vaccinia DNA Topoisomerase I on ice until use.2. Transfer the reactions to a thermocycler or a water bath, incubate at 37℃ for 30 minutes, and then place them on ice.Note: When using a thermocycler for incubation, it is necessary to set the lid temperature at 40℃ to avoid enzyme inactivation.4. Perform electrophoresis with an agarose gel without EB, and stain the gel after electrophoresis for examination.
